THP-1细胞常见问题及解决方法:
1. 培养基里一定要加β-巯基乙醇( β-mercaptoethanol)吗?
β-巯基乙醇是一种常用的培养补充剂,有抗氧化的作用,可减少氧化应激对THP-1细胞的影响。当细胞密度过大但需要维持时,可适当增加巯基乙醇的比例(不超过标准量的1.5倍)。
2. 悬浮细胞在镜下观察时,难以聚焦或估算密度怎么办?
悬浮细胞会随着液体流动,不容易聚焦,静置5-10分钟使细胞沉底,之后再轻轻放上显微镜观察,将有助于聚焦和估算细胞密度。
3.THP-1细胞在培养过程中细胞发生聚团怎么办?
少量细胞聚团,呈葡萄串状,属于正常现象,特别是细胞密度较低时,易出现这种情况。更换血清品牌或增大血清比例(不超过20%)有助于解决聚团问题。不建议将聚团的细胞吹散,等待密度高时,会自己分散开。

4. THP-1细胞在培养过程中细胞发生贴壁怎么办?
少量细胞贴壁属于正常情况,将细胞悬液转移至新瓶中即可。当贴壁细胞的比例高于20%,说明细胞生长环境有异常,需排查培养箱设置、培养基成分,以及培养器皿是否异常。
