UMNSAH/DF-1 is a spontaneously immortalized chicken cell line derived from 10-day-old ELL-0 eggs. Primary chicken embryo fibroblasts were isolated and cultured in medium, passaged until senescence. During senescence, centrifugation was performed to maintain cell cultures at 30%–60% confluency. Non-senescent clones were identified and passaged at least 30 times. No clonal proliferation was observed in soft agar, confirming that these cells are immortalized but not transformed. This cell line can be used as a substrate for virus propagation, recombinant protein expression, and recombinant virus production.
