NIH/3T3 is a highly contact-inhibited, continuous cell line established from the embryonic cultures of NIH Swiss mice. To develop sublines more suitable for transformation analysis in terms of morphological characteristics, the original NIH/3T3 cell line underwent more than five rounds of subcloning. This cell line is highly sensitive to the formation of transformed foci by sarcoma viruses and the replication of leukemia viruses, making it very useful for studies on DNA transformation and transfection. It is negative for ectromelia virus (mousepox virus).
