THLE-2 is an epithelial cell line isolated from the left lobe of a donor. The TTHLE-2 (ATCC: CRL-2706) and THLE-3 (ATCC: CRL-11233) cell lines were derived from normal primary hepatocytes by infection with the SV40 large T antigen. [RF84749] This virus was introduced into the amphitropic packaging cell line PA317 by incorporating the Bgl I-Hpa I fragment of the SV40 T antigen. [RF84750]
THLE-2 and THLE-3 cells exhibit phenotypic characteristics of normal adult liver epithelial cells. When injected into athymic nude mice, they do not form tumors, possess a near-diploid karyotype, and do not express alpha-fetoprotein (AFP). [RF84750] These cells metabolize benzo[a]pyrene, N-nitrosodimethylamine (NDMA), and aflatoxin B1 into their ultimate carcinogenic metabolites, which can bind to DNA, indicating a functional cytochrome P450 pathway. [RF84750] Other enzymes involved in chemical carcinogen metabolism, such as epoxide hydrolase, NADPH-cytochrome P450 reductase, superoxide dismutase, catalase, glutathione S-transferase, and glutathione peroxidase, are also retained in THLE cells.
