MRC-5 is a diploid human cell line composed of fibroblasts, derived from the lung tissue of a 14-week-old aborted white male fetus. The cells exhibit a uniform spindle-shaped morphology.
Using advanced dual transduction technology involving the SV40 large T antigen gene and hTERT (human telomerase reverse transcriptase), Hycyte has successfully developed the MRC-5/TERT-SV40 cell strain, which carries dual resistance to Puromycin (Puro) and Hygromycin (Hygro). This cell line not only retains the classic characteristics of the original MRC-5 cells but also demonstrates excellent passaging capability.

Passaging Capability of MRC-5/TERT-SV40
① Cell condition is sensitive to medium changes. Please use the complete medium provided by Starfish (Product Code: TCH-G619). Using other media may affect cell morphology and passaging ability.
② When using the recommended complete medium and splitting at a ratio of 1:2, we guarantee at least 20 additional passages after receipt.
Trypsinization of MRC-5/TERT-SV40
① This cell line is sensitive to trypsin. Use Trypsin-EDTA (0.25%) with phenol red (NO. GUTP-R002) and incubate at 37°C for 1–2 minutes. Digestion time may vary slightly depending on cell density and brand of trypsin. Digest until approximately 80% of the cells become rounded and begin to detach. Over-digestion (where all cells detach) may occur if left too long, which can negatively impact cell health.
② A small number of floating cells may appear on the second day after passaging. This is normal and can be removed by changing the medium without affecting cell culture.
