This cell line was derived from a pre-treatment sample of a 58-year-old Caucasian male smoker with lung cancer, obtained by A.F. Gazdar, H.K. Oie, J.D. Minna, and their colleagues. It is a hypotriploid human cell line, with the modal chromosome number observed in 68% of counted cells. The incidence of polyploid cells is 3.0%. All cells share 11 marker chromosomes, two of which are paired. The markers include: der(6)(q13), t(7p17q), del(10)(p11.23), the paired der(7)t(7;?17)(p22;?q21.1), and der(16)t(16;?)(q24;?), along with six others. The normal Y chromosome, as well as N13, N15, and N16, are absent; the X chromosome is mostly present in pairs. These cells carry a mutation at codon 191 of the TP53 gene. The cells are unable to synthesize the neuropeptide neuromedin B (NMB) or gastrin-releasing peptide (GRP). They stain positively for keratin and vimentin, but negatively for the neurofilament triplet proteins.
