GP2d and GP5d are derived from the same adenocarcinoma (ECACC catalog number 95090715). These cells originated from a locally recurrent, poorly differentiated Duke's B-stage colon carcinoma surgically removed from a 71-year-old female. The patient had not received any prior chemotherapy or radiotherapy, and she had a strong family history of colon cancer, with two first-degree relatives dying from the disease before the age of 55. The genetic alterations in these two clones are consistent with the tumor progression pattern of colon cancer, yet they exhibit markedly distinct morphologies. A gap deletion on chromosome 5 (region 14q to 22q) and inverted duplications at bands 10q11 and 10q21 have been reported. Both cell lines contain one normal copy of the ki-ras gene and one copy with a mutation at codon 12. GP2d cells show growth responsiveness to EGF, TGF-alpha, or insulin, leading to increased cell numbers. Amphetamine mRNA is abundant in GP2d but nearly undetectable in GP5d.
