SW480 was derived from a primary rectal adenocarcinoma of a 51-year-old Caucasian male patient, while SW620 originated from a lymph node metastasis of the same patient one year later. These cells are negative for CSAp (colon-specific antigen) and rectal antigen 3, but positive for keratin. The p53 gene has a G→A mutation at codon 273 (resulting in an Arg→His substitution) and a C→T mutation at codon 309 (leading to a Pro→Ser substitution). The p53 protein expression level is elevated. The oncogenes c-myc, K-ras, H-ras, N-ras, myb, sis, and fos are expressed positively, whereas N-myc oncogene expression is not detected. The cells do not express Matrilysin (a metalloproteinase associated with tumor invasion). It has been reported that these cells express GM-CSF (granulocyte-macrophage colony-stimulating factor). The ras proto-oncogene is also present.
