RKO is a poorly differentiated human colon cancer cell line. RKO cells contain wild-type p53 but lack the human thyroid hormone receptor nuclear receptor (h-TRβ1). The level of p53 protein in RKO cells is higher than that in RKO-E6 cells. The RKO cell line is the parental cell line for both RKO-E6 and RKO-AS45-1. This cell line is tumorigenic in nude mice and can form colonies in soft agar.
