The EA.hy926 cell line is a human umbilical vein endothelial cell line constructed by fusing primary cultured human umbilical vein cells with a thioguanine-resistant A549 clone under PEG (polyethylene glycol) induction. The hybrid cells were selected and grown in HAT medium, and further screened based on the presence of factor VIII-related antigen. The EA.hy926 cell line has undergone more than 100 population doublings (PDLs). Electron microscopy images show cytoplasmic distribution of Weibel-Palade bodies and tissue-specific organelles, demonstrating differentiated endothelial cell characteristics such as angiogenesis, homeostasis-thrombosis, blood pressure regulation, and inflammatory response.
