A3 subclone is derived from the Jurkat cell line. Jurkat cells were treated with an anti-Fas antibody, and a cell line with a low spontaneous resistance rate to Fas-mediated apoptosis was obtained using the limiting dilution method. The resulting subclone exhibited high sensitivity to Fas-mediated apoptosis. Wild-type A3 cells resistant to neomycin were selected and treated three times with the frameshift mutagen ICR-191 to isolate recessive mutants that were resistant to killing by anti-Fas antibody.
